VONOFast is a next-generation gastric acid suppressant belonging to the Potassium-Competitive Acid Blocker (P-CAB) class. Unlike conventional Proton Pump Inhibitors (PPIs), vonoprazan does not require acid activation and provides rapid, potent, and sustained suppression of gastric acid secretion.
Vonoprazan competitively and reversibly inhibits potassium binding to gastric H+,K+-ATPase (the gastric proton pump) inside parietal cells. Due to its strong basicity, resulting in:
Following administration of vonoprazan for 7 consecutive days:
(Note: While it provides rapid acid suppression from Day 1, any long-term use beyond these timeframes is reserved for maintaining healed reflux or preventing ulcer recurrence in chronic NSAID/aspirin users.)
Concentrations
Gastric Malignancy: Symptomatic response to Vonofast therapy does not preclude the presence of gastric malignancy. Gastric cancer should be ruled out before initiating treatment where clinically indicated
Immediate activity from Day 1 without the need for acid activation.
Maintains target intragastric pH levels for prolonged periods.
Formulated for continuous day and night coverage, with a mean elimination half-life of approximately 6–8 hours.
Flexible once-daily dosing with zero meal timing restrictions.
Rapidly absorbed after oral administration, reaching median peak plasma concentrations (Tmax) within 1.5 hours (fasted) to 3.0 hours (fed). Food has no clinically significant or meaningful effect on overall exposure and absorption
Severe cutaneous adverse reactions, including Stevens-Johnson Syndrome (SJS), Toxic Epidermal Necrolysis (TEN), and Erythema Multiforme, have been reported.
Can be used for a Long-Term Use Continuous administration should be accompanied by regular clinical follow-up.

"Vonofast provides rapid, potent, and sustained acid suppression through innovative P-CAB technology, offering effective management of GERD, peptic ulcer disease, and Helicobacter pylori eradication."
Provides fast onset of action, whereas traditional PPIs demonstrate a slower onset.
Works directly without needing activation in an acidic environment, unlike traditional PPIs.
Provides continuous, powerful acid suppression, whereas traditional PPIs offer variable suppression.
Can be taken with or without food, while traditional PPIs are often heavily affected by meal timing.
Delivers consistent and prolonged pH control compared to the less consistent pH control of standard PPIs.

"In my practice, I frequently treat patients suffering from severe reflux esophagitis. Vonofast has been an excellent addition to our treatment protocols. Because it provides rapid and sustained suppression of gastric acid, my patients consistently report quick relief from their symptoms. Furthermore, the convenience of taking it either with or without food significantly improves patient compliance with the prescribed regimen."
"I regularly treat patients who require long-term non-steroidal anti-inflammatory drugs (NSAIDs), which unfortunately puts them at a high risk for developing ulcers. I often prescribe Vonofast 10 mg once daily as a preventative measure. It has proven highly effective in preventing the recurrence of gastric and duodenal ulcers associated with NSAID administration, giving me peace of mind that my patients' digestive health is protected."
"Vonofast is a highly reliable potassium-competitive acid blocker (P-CAB) that I frequently dispense for patients undergoing Helicobacter pylori eradication therapy. When used as an adjunct therapy alongside antibiotics like amoxicillin and clarithromycin, it plays a vital role in the treatment's success. I always make sure to counsel patients that the film-coated tablets must be swallowed whole and should not be chewed or crushed to ensure the medication works as intended."
"Managing patients who experience a recurrent relapse of healed reflux esophagitis can be challenging, but maintenance therapy with Vonofast has yielded fantastic results in my clinic. The 10 mg daily dose is highly effective for long-term management up to 6 months. Its pharmacokinetic profile is impressive, generally reaching peak plasma concentrations within just a few hours after oral administration, which allows for prompt and dependable acid suppression."
